Difficulties with conventional phytoplasma diagnostic using PCR/RFLP analyses
نویسنده
چکیده
Polymerase chain reaction (PCR) with subsequent restriction fragment length polymorphism (RFLP) analysis is often used for phytoplasma identification and classification. Although these techniques are very sensitive and specific, in some cases, nonspecific reactions, false positives and negatives results, as well as unusual or illegible profiles after RFLP analyses, amplification of plant host ́s DNA or other difficulties occurred. Experiences with suitability of positive and negative controls integration in PCR, evaluation of critical samples and other difficulties in phytoplasma PCR/RFLP identification are reported.

Classification of new phytoplasmas associated with diseases of strawberry in Florida, based on analysis of 16S rRNA and ribosomal protein gene operon sequences.
Strawberry plants exhibiting symptoms of stunting and abnormally small leaves were observed in production fields in central Florida, USA. Since the symptoms were suggestive of phytoplasma infection, plants were assayed for presence of phytoplasma by PCR amplification of 16S rDNA and ribosomal protein (rp) gene sequences. Amplification of phytoplasma-specific DNA sequences by PCR indicated infec...
متن کاملPhytoplasma classification: Taxonomy based on 16S ribosomal gene, is it enough?
The discovery in 1967, of a new group of plant pathogens related to bacteria led to the finding of pleomorphic, wall-less prokaryotes in the phloem of many plant species affected by yellows-type diseases. Following the application of molecular technologies the phylogeny of these prokaryotes was resolved and led to the new trivial name of “phytoplasma” and to the designation of a new taxon named...
متن کاملMolecular and symptom analyses of phytoplasma strains from lettuce reveal a diverse population.
ABSTRACT Epidemics of aster yellows in lettuce in Ohio are caused by at least seven distinct phytoplasma strains in the aster yellows (AY) group. Five of the strains are newly reported: AY-BW, AY-WB, AY-BD3, AY-SS, and AY-SG. All seven strains were characterized based on symptoms in aster and lettuce, and by polymerase chain reaction (PCR). Strain AY-BD2 (formerly 'Bolt') causes yellowing and l...
متن کاملCharacterization and vector identification of phytoplasmas associated with cucumber and squash phyllody in Iran
Phytoplasmas associated with cucumber phyllody (CuP) and squash phyllody (SqP) in Yazd province of Iran were characterized by molecular analyses and biological studies. Orosius albicinctus leafhoppers testing positive for phytoplasma presence by polymerase chain reaction (PCR) successfully transmitted CuP and SqP phytoplasmas to healthy cucumber and squash plants. The phytoplasmas were also tra...
متن کاملMolecular characterization, vector identification and sources of phytoplasmas associated with brinjal little leaf disease in India
Brinjal little leaf (BLL) is a widespread disease of phytoplasma etiology in India that induces severe economic losses. Surveys were conducted in eight brinjal-growing states of India during July 2014 to September 2015 and eighteen BLL samples showing little leaf, phyllody and witches' broom symptoms were collected for phytoplasma identification. Presence of phytoplasmas was confirmed in all th...
